When the ISO 5459 is active globally, (Ref. Calculation as per ISO 5459 & Outer Tangential Element are checked), will this only constrain the datums correctly when adding them individually into position & profile?
Or when I create any secondary print origin alignments or (geometry best fit alignments), and use an alignment(s) in the DRF drop down box in the position or profile characteristic, will this active the (ISO 5459)? Or would I need to constrain the alignment(s) manually in the alignment template itself?

No ISO knowledge here, but in my experience it's best to construct your own alignment then reference it in the characteristic. This gives you more control over how the DRF is assembled, where as selecting individual features in the characteristic leaves it up to Calypso (and its logic) and follows the Can / May / Must rule (if a datum CAN restrict a degree of freedom, and is PERMITED to restrict a degree of freedom, then it MUST restrict a degree of freedom.) But, your question is whether or not your ISO standard stays applicable when building your own DRF or letting Calypso do it; that, I'm not sure.

Yes, to the first question.

No, to the second question. If you build secondary alignments, you would have to manually convert the features to Outer Tangential Element and Constrain per ISO 5459.
